Abstract
Function of the estrogen hormone is intimately correlated and regulated by the estrogen receptor alpha (ER-alpha) gene. Estrogen hormone influences the development and function of reproductive organs, such as the mammary gland, ovary, and uterus. In this current research, we cloned and characterized a partial coding sequence (CDS) of exon 1 from the ER-alhpa gene in Sahiwal and Hariana cattle breeds and study investigating polymorphisms was carried out in 5’ UTR of ER-alpha gene using BglI/PCR-RFLP assay. The nucleotide and amino acid sequences of the ER-alpha exon 1 in Sahiwal and Hariana cattle were highly analogous to those in exotic and crossbred cattle breeds. The PCR-RFLP assay uncovered three different genotype categories namely AA, AB, and BB where genotype AB was almost as frequent as BB genotype followed by AA genotype and the frequency of the B allele was higher than that of A allele in all the screened animals. Chi-square analysis indicated that the examined animal population adhered to the Hardy-Weinberg equilibrium. While an association study detected a notable significant (p<0.05) difference in lactation I where the BB genotype showed a higher lactation period than other genotypes in Hariana cattle. Future research, to identify the function of the cattle ER-alpha gene as a potential gene marker, will be strengthened by the information provided from our study. Our results should be further validated with a larger sample size that can be later implemented in the upgradation of the breeding programs of the studied cattle breeds.
